4Cell® SmartCHO media 는 최신의 세포공학 기술에 의해 최근 새로이 개발된 media 로서 모든 종류의 CHO cell line 에 사용할 수 있는 가장 탁월한 세포 배양 성능을 가진 media & feed system 입니다.

fed-batch application 용으로 사용되는 4Cell® SmartCHO media 는 다양한 종류의 CHO cell line 에 대해 테스트를 진행한 결과 모든 CHO cell line 의 성장에 탁월한 성능을 보였으며 또한 mAB, bi-specifics, 그리고 Fc fusion proteins 와 같은 product 의 생산 효율이 매우 우수하였습니다.

CHO-GD44 cell line 을 이용하여 4Cell® SmartCHO media 와 다른 유명 브랜드 media 의 fed-batch cilture performance 를 비교하는 테스트를 진행하였습니다. 또한 CHO-S, CHO-K1, CHO-GS cell line 도 추가로 이 비교테스트에 사용하였으며 세포배양은 Sartorius 의 Ambr 15 microbioreactor 를 이용하였습니다.

테스트 결과 4Cell® SmartCHO media 는 다른 유명 브랜드 media 보다 CHO cell 배양 능력이 더 우수하다는 것을 확인할 수 있었습니다.

Peformance Test


아래 Figure 1 과 Figure 2 는 4Cell® SmartCHO media 의 CHO cell line 을 배양하는 능력이 다른 유명 브랜드 media 에 비해 일부분은 유사하지만 대부분 훨씬 더 탁월하다는 실험 결과를 보여주고 있습니다.

Figure 3 은 CHO 세포의 특정 생산성은 클론과 배지에 따라 변동성이 매우 큰데 이는 cell line 과 응용 분야에 따라 다양한 formulations, feeds, 그리고 parameter 들을 조사할 필요성이 있다는 것을 이야기하고 있습니다.

cell culture media 는 세포가 생산하는 product 의 품질에 매우 큰 영향을 미친다. Size-exclusion chromatography 테스트 결과 CHO-S 세포는 약간의 차이는 있지만 media 와 상관없이 매우 낮은 수준의 low aggregate values 를 유지하였습니다. CHO-DG44 (clone 1) 의 경우에는 4Cell® SmartCHO media 에서 가장 낮은 aggregate values 를 나타내었습니다.

Cell culture media 의 성능은 대용량 배용에서도 일정하여야 합니다. Figure 5 의 데이터에서 우리는 4Cell® SmartCHO media 를 5 리터 bioreactor 에서의 배양에 사용한 결과 세포 배양 결과가 소용량때와 동일하고 배양도 매우 탁월하였다는 것을 알 수 있습니다.
