VELP Sensor System ÀÇ Respirometric Sensor System À» Ȱ¿ëÇϸé BMP (Biochemical Methane Potential, »ý¹°ÇÐÀû ¸Þź »ý¼º °¡´É¼º) ¸¦ Á¤·®ÀûÀ¸·Î ÃøÁ¤ÇÒ ¼ö ÀÖ½À´Ï´Ù.

Çø±â¼º ºÐÇØ´Â »ê¼Ò°¡ ¾ø´Â »óÅ¿¡¼­ º¹ÀâÇÑ À¯±â¹°ÁúÀÌ ºÐÇØµÇ¾î ¸Þź°ú ÀÌ»êȭź¼Ò·Î ±¸¼ºµÈ °¡½º¸¦ »ý¼ºÇÏ´Â »ýÈ­ÇÐÀû °úÁ¤ÀÔ´Ï´Ù. ÀÌ °úÁ¤Àº ƯÁ¤ Çø±â¼º ¹Ì»ý¹°¿¡ ÀÇÇØ ¹ß»ýÇÕ´Ï´Ù. BMP ÃøÁ¤Àº, ÀÏÁ¤·®ÀÇ ±âÁú¿¡ Çø±â¼º ¹Ì»ý¹°À» Á¢Á¾Çϰí, ÀÏÁ¤ÇÑ ¿Âµµ (¿¹: 35?¡ÆC) ¿Í ±³¹Ý Á¶°ÇÀ» À¯ÁöÇÑ »óÅ¿¡¼­ ÀÌ·ç¾îÁý´Ï´Ù. ¿ë±â´Â ¹«»ê¼Ò ºÒȰ¼º °¡½º È¥ÇÕ¹° (¿¹: Ar) À» ÁÖÀÔÇÏ¿© Çø±â¼º Á¶°ÇÀ¸·Î ÀüȯµË´Ï´Ù. ½ÃÇèÀÌ ½ÃÀÛµÇ¸é ºÐÇØ °úÁ¤ÀÌ ÁøÇàµÊ¿¡ µû¶ó ¿ë±â ³»ºÎ¿¡¼­ ¹ÙÀÌ¿À°¡½º°¡ »ý¼ºµÇ¸ç, ¹ÙÀÌ¿À°¡½º·Î ÀÎÇØ ¿ë±â ³»ºÎÀÇ ¾Ð·ÂÀº Áõ°¡ (overpressure) ÇÏ°Ô µË´Ï´Ù.

¸Þź°ú ÇÔ²² »ý¼ºµÈ ÀÌ»êȭź¼Ò´Â ¿ë±â »ó´Ü¿¡ À§Ä¡ÇÑ °­¾ËÄ®¸®¼º ¹°Áú (KOH Æç·¿) ¿¡ ÀÇÇØ Á¦°ÅµÇ¾î, °á°ú´Â mLCH4@STP/gVS ·Î ³ªÅ¸³ª°Ô µË´Ï´Ù. ÀÌ´Â Èֹ߼º °íÇü¹°·Î ÀüȯµÈ À¯±â¹° ´ÜÀ§´ç Ç¥ÁØ »óÅ¿¡¼­ »ý¼ºµÈ ¸ÞźÀÇ ºÎÇǸ¦ ÀǹÌÇÕ´Ï´Ù.

ºÐ¼®Àº ÀϹÝÀûÀ¸·Î 37¡ÉÀÇ ¿Âµµ Á¶°Ç¿¡¼­ 30ÀÏ ÀÌ»ó ÁøÇàµÇ¸ç, ½ÃÇè Á¾·á ±âÁØÀº 3ÀÏ ¿¬¼ÓÀ¸·Î ÃøÁ¤µÈ ÀÏÀÏ ¸Þź »ý»ê·®ÀÌ ´©Àû ¸Þź »ý¼º·®ÀÇ 1% ¹Ì¸¸ÀÏ °æ¿ì, ½ÃÇèÀ» Á¾·áÇÕ´Ï´Ù.

´ÙÀ½Àº Respirometric Sensor SystemÀ» ÀÌ¿ëÇÑ BMP ÃøÁ¤ ÀýÂ÷ÀÔ´Ï´Ù
1. Set the incubator temperature to the desired value, e.g., 35¡ÆC
2. Connect the Wireless DataBoxTM and fill in the RESPIROSoftTM software database
3. Introduce a magnetic stir bar into each bottle
4. Weigh precise the mass of well mixed inoculum in a beaker
5. Add the nutrients solutions by cylinder
6. Add the substrate, cellulose, (except for the ¡°blank¡± bottles where this volume is replaced by an equivalent volume of dilution water).
7. Check the pH of the mixture and, if lower than 7.3, adjust it (when removing the pH probe from the mix, be careful not to remove too much material from the bottle).
8. Flush the headspace with nitrogen (or argon) deoxygenated gas: according to the flow for 5 to 10 minutes
9. Introduce about 20 g of KOH into the alkali collector, below the holes.
10. Screw the RESPIROMETRIC Sensors onto each bottle and tighten.
11. Place the system in the thermostat at the desired temperature, without stirring.
12. Wait about an hour for the liquid mass present in the bottle to reach the test temperature, and then to develop the typical initial pressure increase, due to the vapor pressure. To remove this additional pressure, it is advisable, after one hour, to vent the test bottles to atmospheric pressure (by briefly loosening the ring nut).
13. The system is ready to start the BMP measurement. Press START and start stirring.
